三方抑制剂插入到NK1R附近(1)跟随NK1R一起内吞并在细胞内抑制NK1R激动剂与NK1R的结合从而抑制疼痛信号的传递(2)
也就是说,之前科学家们之所以失败,是因为他们没有想到NK1R还“留了一手”,原来细胞内才是传递疼痛信号的主要场所。对于这一发现的意义,大家应该也很清楚了。在美国,有超过5000万成年人正遭受慢性疼痛的折磨(14),而由于我国庞大的人群基数,在我国慢性疼痛患者可能更多,有研究显示仅在香港,成年人中慢性疼痛患者就超过3成(15)。所以Bunnett教授的这一发现无疑将会为慢性疼痛患者带来了新的治疗选择,有望减少甚至替代阿片类镇痛药的使用。
