• 乳腺癌易感基因包括:ATM, BARD1, BRCA1, BRCA2, CDH1, CHEK2, PALB2, RAD51C & RAD51D
• 在临床检测和基于人群的队列中,分别有6.5%和4.6%的DCIS女性携带种系致病变异(PV)
• 相比于浸润性导管癌(IDC)女性,DCIS女性的BRCA1、BRCA2和PALB2 PV的发生率显著更低(临床队列:2.8% vs. 5.7%;基于人群的队列:1.7% vs. 3.7%),而ATM和CHEK2的PV发生率相似
• 两个队列中,ATM、BRCA1、BRCA2、CHEK2和PALB2 PV与DCIS风险增加显著相关(OR>2.0),但只有BRCA2 PV与高危(OR>4)相关
• 在DCIS高渗透性基因的PV携带者中,对侧乳腺癌的15年内累积发病率为23%
